摘要 采用根癌农杆菌介导DREB—like转录因子基因真空渗入浸染转化烟草,结果表明:真空恢复常压时间的长短是影响转化效率的主要因素之一。以恢复常压时间为180s时,转化效率最高(78%);恢复常压时间为50s时,叶盘水渍严重,转化率仅有23%;恢复常压时间为360s时,转化率下降为68%。真空渗入浸染与普通浸染相比转化率高,出芽提早2~3d,再生芽健壮,生长快,但0.04MPa和0.08MPa2个真空度下浸染处理,转化率没有大的差异。 The transformation of leaf segments of tobacco mediated by Agrobacterium tumefaciens LBA4404 with plasmid pROK2 by vacuum infiltration infection showed that the time span from vacuum to natural atmosphere was a driving factor affecting transformation efficiency. Transformation rate was 78% when the time was 180s and dropped to 68% at 360s, 23% at 50s. Vacuum infiltration seemed to promote transformation, resulting in shoot emerging earlier 2 or 3 days, and growing rapidly and strongly, but it had no effects on the transformation efficiency at 0.04MPa and 0.08MPa.
