摘要
小麦穗发芽与内源α-淀粉酶活性的提高密切相关,抑制内源α-淀粉酶活性是解决小麦穗发芽问题的关键。本研究对11份普通小麦以及1份人工合成抗穗发芽六倍体小麦RSP的α-淀粉酶抑制因子基因,进行了分离克隆与序列测定,并与核酸数据库中已知序列进行比对分析发现,α-淀粉酶抑制因子基因相当保守,一致性达到94.83%,仅在少数位置出现单碱基的替换或单个碱基的插入。在22份对比材料中,编码序列的229、272、397、415、427、430 bp位置处,发生频率为9.1%的单核苷酸替换,是潜在的单核苷酸多态性(SNP)位点。
The pre-harvest sprouting of wheat was determined by the activity of α-amylase. It is important to reduce the activity of α-amylase to control the pre-harvest sprouting. The α-amylase inhibitor genes, which were related to the pre-harvest sprouting resistance, were isolated from 11 common wheat varieties and 1 synthetic wheat line RSP. Sequence analysis indicated that the α-amylase inhibitor genes were highly conserved and they shared very high coherence (94.83%) with the insertion or deletion of single nucleotide in a few locations. At positions 229, 272, 397,415, 427 and 430 bp of the nucleotide sequence, the candidate single nucleotide polymorphisms (SNPs) were observed with the frequency of 9.1%.
