摘要 目的:用地塞米松磷酸钠(DSP)乳酸羟基乙酸共聚物(PLGA)微球干预内毒素(LPS)所致大鼠急性肺损伤(ALI),探索其对ALI的保护作用。方法:复制大鼠ALI模型,随机分为4组:空白组(C)、内毒素组(L)、地塞米松磷酸钠组(D)、DSPPLGA微球组(M)。观察肺组织病理改变、肺湿干重比(W/D)、支气管肺泡灌洗液(BALF)中PMN百分比及血中IL8和IL10浓度的变化。结果:①LPS致大鼠急性肺损伤模型成功;②大鼠肺病理半定量评分,肺W/D,血IL8浓度和BALF中PMN百分比,L组明显高于其它3组,D、M两组间无显著性差异;③血IL10浓度,L组升高与C组有显著性差异,与D、M两组无显著性差异,且D、M组间无显著性差异。结论:DSPPLGA微球对ALI有保护作用。 Objective: To investigate the protedive effect of microspheres that contained dexamethasone sodium phosphate (DSP)encapsulated by the biodegradable synthetic polymers PLGA on murines of acute lung injury (ALl) induced by LPS. Methods: A rat model of ALl was induced by LPS. Twenty-four rats were randomly divided into four groups: normal control group (C),LPS group (L),DSP group (D) and microspheres group (M). Histopathological examinations and the wet-to-dry lung weight ratio (E/D) were recorded. The percentage of PMN in mice bronchoalveolar lavage fluid (BALF) and IL-8 and IL-10 in mice plasma were examined. Results: ① Pathological examination with light microscope in LPS group showed thickened pulmonary interstitial, inflammatory cell infiltration, edema and hemorrhage, that is to say the model of ALl were established successfully. ② The grades of ALI pathologic change, W/D in the lung, IL-8 in the blood and PMN% in the BALF in LPS group were significantly higher than those of the other groups, which were significantly decreased in DSP group and microspheres group as compared with those of LPS group (all P〈0.05), but no significant difference between DSP group and microspheres group (all P〉0.05) was found. ③There was significant difference in the concentration of interleukin-10 among the four groups (P〈0.01). The concentration of IL-10 in LPS group was significantly higher than that of the control group (P〈0.01) and was not significantly different to that of DSP group and microspheres group (both P〉0.05). And no difference between DSP group and microspheres group (P〉0.05) was found. Conclusion: The DSP-PLGA microspheres could protect ALI induced LPS.
