摘要
目的探讨情绪应激对NOD小鼠脑内c-fos表达和对胰岛内细胞凋亡的影响,了解应激影响糖尿病发生的中枢神经机制。方法自发糖尿病模型NOD小鼠在接受饮水冲突应激加限制应激后,分别于接受应激1天、2天、3天时,原位杂交测定脑区c-fos表达水平,TUNEL法测定胰岛细胞凋亡水平。结果1天(3.93±1.05)、2天(5.73±2.60)、3天(5.77±2.75)时段应激组小鼠胰岛细胞凋亡发生率显著高于对照组(0.82±0.93,t=4.46、3.58、3.41,P<0.05)。应激引起c-fos mRNA在脑内的广泛区域表达,其中海马三区、杏仁中央核、下丘脑室旁核的c-fos mRNA表达增高与胰岛细胞凋亡增高间存在正相关(0.79、0.69、0.64,P<0.01或P<0.05)。结论情绪应激时c-fos在海马三区、杏仁中央核、下丘脑室旁核中表达的提高与胰岛细胞凋亡升高相关,提示它们可能参与了应激影响1型糖尿病发生的过程。
Objective: To explore the effect of emotional stress to the intra-islet apoptosis and the it's neuralmechanism. Methods: Using the drinking-conflict stress and restraint stress, we investigated diabetes-prone NOD mouse's c-fos expression in it's brain and it's intra-islet apoptosis. Correlation between the two variables was alsostudied. Results: The average intra-islet apoptosis of the first ( 3.95±1.05 ) and the second ( 5.73±2. 60 ) andthird day ( 5.77±2. 75 ) was higher than the control's (0. 82±0. 93, P 〈 0. 05 ) . c-fos mRNA expression in cornuammonis, central amygaloid nuclei and hypothalamic paraventricular nucleus after stress was correlated to the intra-is-let apoptosis (r =0. 79, 0. 69, 0. 64 ; P 〈 0. 01 or P 〈 0. 05 ) . Conclusions: The correlation between intra-islet apoptosis and c-fos mRNA expression in cornu ammonis, central amygoloid nuclei and hypothalamic paravertricular nucleusafter stress indicated that these brain region may contribute to onset of type 1 diabetes after stress.
