摘要
目的:观察雌激素对κ阿片肽受体(κOR)mRNA含量和蛋白表达的调节作用.方法:采用半定量RT—PCR和Western blot方法,检测17β-雌二醇作用24~72h后CNE2细胞κOR mRNA含量和蛋白表达的变化.结果:17β-雌二醇(μmol/L)作用24h后,CNE2细胞表达κOR mRNA和蛋白明显降低,mRNA和蛋白含量分别下降到对照组的(49.2±7.66)%(P〈0.01)和(60.8±8.36)%(P〈0.01);作用时间延长到48h后,CNE2细胞κOR蛋白含量为对照组的(32.9±7.13)%(P〈0.01);而延长到72h后,κOR mRNA和蛋白含量分别下降到对照组的(20.8±7.71)%(P〈0.01)和(29.8±8.41)%(P〈0.01).结论:雌激素能够从转录水平抑制κOR在CNE2细胞的表达.
AIM: To observe the regulatory effect of 17β- estradiol on the expression of kappa opioid receptor (KOR) mRNA and protein. METHODS: Semi-quantitative RT-PCR and Western blot were used to analyze the mRNA content and protein expression in CNE2 cells 24 -72 h after treated with 17β-estradiol. RESULTS: Compared with that in control group, KOR mRNA reduced to (49.2±7.66)% (P〈0.01) and (20.8 ± 7.71 ) % ( P 〈0.01 ) respectively, after 1 μmol/L 17β-estradiol treatment for 24 and 72 h. The quantity of protein decreased to (60.8±8.36)%, (32.9 ±7. 13)% and (29.8 ±8.41)% (P 〈0.01 ) respectively following 1 μmol/L 17 β-estradiol treatment for 24, 48 and 72 h. CONCLUSION : Estrogen can inhibit the transcription of KOR in CNE2 cells.
