摘要
用λ噬菌体载体构建了条斑紫菜Porphyra yezoensis丝状体cDNA表达文库,并对文库进行了随机测序,对随机测序形成的719条表达序列标签(EST)序列进行了初步分析。结果表明:该文库包含1.52×106个独立克隆,重组率为88.2%;EST的冗余率为54.2%,新EST的比率为65.7%。本研究为开展条斑紫菜的功能基因克隆、分析和功能基因组学研究提供了条件。EST序列已递交Gen-Bank,编号为CX873884到CX874602。
A laver Porphyra yezoensis sporophyte cDNA expression library has been constructed with λ phage vector and was randomly sequenced. The library consists of 1.52 × 10^6 clones with the recombinant percentage of 88.2%. The analysis of the 719 ESTs showed that the redundancy was 54.2% and the proportion of new EST was 65.7%. This library can be used as the base for gene cloning and analyzing, and further for functional genomic research of the laver. The nucleotide sequence data reported has been deposited in GenBank under accession nos. from CX873884 to CX874602.
