兔动脉粥样硬化狭窄模型制备及评价方法研究

摘要 目的 制备动脉粥样硬化狭窄的模型,用超声和血管造影进行评价动脉狭窄程度。并比较一致性。方法 雄性新西兰大白兔(体重2.5~3.5kg,4月龄)10只,用股动脉穿刺法行腹主动脉内膜剥脱术同时饲喂含1.5%胆固醇的高脂饮食,于术后第9周行腹主动脉超声和血管造影检查,记录腹主动脉狭窄程度。结果 第9周时腹主动脉超声检查均显示血管狭窄(51.11%±5.77%vs57.00%±7.89%,P〉0.05),两者的相关性r=0.89。结论 动脉内膜剥脱术同时饲喂高胆固醇饮食可以在短时间内复制出动脉粥样硬化狭窄模型,超声和血管造影均能准确判定血管狭窄程度。 Objective To make models of atherosclerosis rabbits , to evaluate the degree of stenosis of abdominal aorta by ultrasonographic image and angiography and to compare the consistency of the two measures. Methods Male New Zealand rabbits ( n = 10, weight 2.5 - 3.5 kg,4 month old) were fed a cholesterol- supplemented diet( 1.5% )and were subject to balloon catheter injury of the abdominal aorta by femoral artery. Ultrasonographic images were performed in 9th week after balloon denudation. After that , angiography on abdominal aorta was recorded by DSA. Results Stenosis of abdominal aorta were observed in 9th week by both uhrasonographic image and arteriography(51.11 ± 5.77% vs 57.00 ± 7.89% , P 〉 0.05 ), the correlation between them is 0.89. Conclusion The animal model of stenosis could be successfully established in a short time by balloon endothelial denudation of abdominal aorta in cholesterol -fed rabbits. Doppler ultrasonography appear suitable for measuring the degree of stenosis of the abdominal aorta when compared to angiography.
