烟碱降解菌的选育及改善上部烟叶品质研究 被引量:24

摘要 从烟叶叶面和植烟土壤中分离到降解烟碱的微生物18株,经初筛和复筛得到降解烟碱活性较高的微生物菌Nic22。对该菌株产酶培养条件进行优化研究,实验表明:Nic22降解烟碱效果受到多种因素的影响,添加适量的精制淀粉和(NH4)2SO4作为外加碳氮源可明显促进烟碱的降解,适宜的摇瓶培养温度、pH和时间分别为30℃、6.0~7.0和9h。采用Nic22菌株所产酶液对上部烟叶进行处理试验,卷烟评吸结果表明:上部烟叶经酶液处理可明显减轻杂气和刺激性,抽吸品质得到显著提高。 18 bacterial strains were screened from tobacco leaves and soil of tobacco field. The microorganism Nic22 with high nicofine-deoomposing activity was obtained by repeated screening. The culture conditions for enzyme production were optimized The addition of refined starch and (NH4)2SO4 could promote the ability of nicotine degradation. The optimum of temperature, pH and time of fermentation were 30℃, 6.0 - 7.0 and 9h, respectively. The upper leaf of flue-cured tobacco was treated with the crude enzyme from strain Nic22. The results showed that the enzyme treatment of the upper-tobacco leaf could reduce irritation and improve the smoking quality significantly.
