摘要
以3个烤烟品种为试验材料,对不同陈化时期烟叶中多酚氧化酶、过氧化物酶活性进行了分析,同时利用电泳技术研究了不同陈化时期烟叶中多酚氧化酶和过氧化物酶同工酶酶谱的变化。结果表明随着陈化时间的延长,多酚氧化酶、过氧化物酶活性呈现先升高后降低的趋势;多酚氧化酶和过氧化物酶同工酶酶谱在陈化过程中发生了较为显著的变化,表现为陈化3个月以前酶带较少,酶带的亮度较弱,陈化6个月时酶带最多,亮度最强,6个月以后酶带数量开始减少,亮度也有所下降。通过酶谱分析,也进一步验证了不同陈化时期烟叶中多酚氧化酶和过氧化物酶活性变化规律。
The activities of polyphenol oxidase and peroxidase in three flue-cured tobacco varieties during aging were analysed and the changes of the zymograms of the two enzymes' isozymes were studied parallelly with electrophoresis. The results showed that the activities of polyphenol oxidase and peroxidase increased first and then decreased in the course of aging; the zymograms of the isozymes of polyphenol oxidase and peroxidase changed relatively remarkably during aging. It demonstrated that: during the first three months of aging, only a few dim isozyme bands were presented; the amount and brightness of isozyme bands peaked in the 6th month of aging, and diminished gradually afterwards. The analysis of the zymograms of the isozymes verified the rule of activity change of oxidase and peroxidase in tobacco leaf during aging.
