摘要
目的研究白芍总苷(TGP)对胶原性关节炎滑膜组织中Bcl2、Bax表达的影响。方法SD大鼠随机分成4组,正常组、模型组和TGP大、小剂量组;采用鸡Ⅱ型胶原皮内注射诱导大鼠胶原性关节炎(CIA)模型;运用免疫组织化学法检测各组滑膜组织Bcl2、Bax蛋白的表达。结果正常SD大鼠滑膜组织Bcl2和Bax均有表达,模型组Bcl2、Bax表达明显高于正常组,但Bcl2/Bax比例比正常组高;TGP(25、50mg/kg)治疗组可明显下调Bcl2蛋白的表达,上调Bax蛋白表达,Bcl2/Bax比例下降,与模型组比较,差异有显著性(P<0.01)。结论Bcl2、Bax参与CIA的发病过程,TGP可下调滑膜组织中Bcl2表达、上调Bax表达及降低Bcl2/Bax的比值,这可能是其治疗CIA的机制之一。
Objective To investigate the effect of total glucosides of paeony(TGP)on Bcl-2, Bax expression in rats of synovial tissue with collagen-induced arthritis(CIA). Methods SD rats were divided randomly into four groups including normal, model, TGP(25, 50 mg/kg) groups. Chicken type Ⅱ collagen-induced arthritis model was established in rats. The expression of Bcl-2, Bax on synovial tissue were detected by means of immunohistochemistry. Results The expression of Bcl-2, Bax was higher in model group than that of the normal group. And compared with the model group , the expression of Bcl-2 protein was decreased, Bax protein was increased and the ratio of Bcl-2/Bax was decreased in TGP groups. Conclusion Bcl-2 and Bax protein play a role in the development of CIA. TGP can decrease the expression of Bcl-2 and increase the Bax protein ,then decrease the ratio of Bcl-2/Bax, which may be one of the mechanism of TGP improvement CIA.
