摘要
在临床ＰＣＲ检验中，血卟啉对酶促反应的影响不容忽视，因此在纯化ＤＮＡ的过程中，常需将红细胞清除，以消除血卟啉对实验结果的干扰。用ＴＫＭ法提取血液ＤＮＡ，方便、经济、快捷且效果良好。该ＤＮＡ可用于ＰＣＲ的分析，结果极为理想，完全可以替代其他血液ＤＮＡ的提取方法。
new method for DNA preparation from whole blood-TKM was reported. Usually, the blood was digested with proteinase K after gradient centrifugation,then DNAwas extracted wi th phenol and chloroform. With the new method-TKM, a low ionicconcentration solution TKM1 was only needed to destroy the red blood cell. As a result,theprocess of DNA preparation became easy and simple. And what is more, there was no sig-nificant difference in the purity of DNA between the TKM and the previous method(P>0. 05). In the clinical PCR tests,the enzymic activity had not been interfered byporphyrin.
