摘要
目的研究皮肤鳞状细胞癌(SCC)和基底细胞癌(BCC)中缺氧诱导因子-1α(HIF-1α)的表达,分析肿瘤组织中HIF-1α与血管生成的相关性。方法采用免疫组化S-P法检测石蜡包埋组织(其中SCC 25例,BCC 25例,正常皮肤20例)中HIF-1α的表达,并用CD34标记血管内皮细胞,计算微血管密度(MVD)。结果HIF-1α主要表达于瘤细胞胞浆中,在两种肿瘤组织中均呈弥漫性表达;CD34表达于血管内皮细胞胞膜,分布于间质及癌巢。HIF-1α在SCC和BCC中表达阳性率分别为88%(22/25)、56%(14/25),SCC与BCC比较有显著性差异(P<0.05)。HIF-1α在正常皮肤中阴性表达。MVD平均值在SCC和BCC中分别为42.55±5.02,23.47±6.21,两者比较有显著性差异(P<0.05)。HIF-1α阳性的肿瘤组织MVD高于其表达阴性者(P<0.05)。结论HIF-1α在两种肿瘤组织尤其是SCC中过表达,HIF-1α可能通过促进肿瘤新生血管形成,从而促进SCC和BCC的生长、浸润和转移。
Objective To investigate the relationship of the expression of hypoxia-inducible factor-1α (HIF-1α) with angiogenesis in cutaneous squamous cell carcinoma (SCC) and basal cell carcinoma (BCC). Methods The expression of HIF-1α was detected by immunohistochemistry (S-P) in 25 paraffin-embedded specimens of SCC, 25 BCC and 20 normal skin. CD34 immunostain was used to measure the microvascular density (MVD). Results Distribution of HIF-1α was demonstrated within cytoplasm of tumor cells, and diffusive distribution was in both kinds of tumors; distribution of CD34 was demonstrated within cytomembrane of vascular endothelial cells, and located in the stroma around the nests and the tumor nests. The positive expression rates of HIF-1α protein in SCC, BCC were 88 % (22/25), 56% (14/25) respectively. There was a significantly difference in the expression of HIF-1α between SCC and BCC (P 〈 0.05). The expression of HIF-1α in the normal skin was negative. MVD was much higher in tumor with HIF-1α ( + ) than in tumor with HIF-1α ( - ) ( P 〈 0.05). Conclusions There are overexpressions of HIF-1α in both kinds of tumors especially in the SCC. HIF-1α in tumor is correlated with the neovascularization of the tumor, so it may promote tumor development, invasion or metastasis.
