摘要 目的:研究人胰腺癌细胞株和胰腺癌组织中KiSS-1表达及其临床意义。方法:应用RT-PCR方法检测KiSS-1基因mRNA在人胰腺癌细胞株AsPC-1、40例胰腺癌及27例转移灶和9例正常胰腺组织中的表达情况。结果:人胰腺癌细胞株AsPC-1和胰腺癌组织中KiSS-1mRNA表达水平均显著低于正常胰腺组织(P值均为0·000),其中As-PC-1中表达水平最低,为0·124±0·046。KiSS-1mRNA的表达与性别、年龄、肿瘤大小及组织学分级无关,与临床分期、有无转移和神经侵犯密切相关。在Ⅲ、Ⅳ期胰腺癌组织中KiSS-1基因的表达为0·137±0·110,明显低于Ⅰ、Ⅱ期,P=0·000。KiSS-1基因在无转移和有转移的胰腺癌原发灶和转移灶组织中表达水平呈降低趋势,差异有统计学意义(P值均为0·000),转移灶中的表达水平最低。有神经侵犯的胰腺癌原发灶KiSS-1基因表达显著低于无神经侵犯的胰腺癌原发灶(P=0·000),其表达与有无神经侵犯Kendall等级相关系数τ=0·689,P=0·004。结论:KiSS-1基因转录和表达降低与胰腺癌侵袭和转移密切相关,可能参与胰腺癌侵袭和转移的调控。 OBJECTIVE: To investigate the expression of KiSS-1 in human pancreatic cancer cell line and tissues and its clinical significance. METHODS: RT-PCR analysis was used to detect the expression level of KiSS-1 mRNA in human pancreatic cancer cell line AsPC-1 and tissues of 40 cases of pancreatic cancer, 27 cases of their metastastic site and 9 cases of normal pancreatic tissues. RESULTS: The mRNA expressions of KiSS-1 in AsPC-1 cells and pancreatic tissues were significantly lower than those in normal (P=0. 000 ,P=0. 000), which was the lowest in AsPC-1 cell (0. 124±0. 046). The expression of KiSS-1 had not correlation with sex, age, size of tumor and cell differentiation. However, the expression of KiSS-1 was related to the clinical stage, metastasis and nerves invasion significantly. The expression of KiSS-1 mRNA was 01 137 ± 0. 110 in stage Ⅲ and Ⅳ, which was significant lower than that in stage Ⅰ and Ⅱ , P = 0. 000. It showed a decreased tendency in pancreatic cancer without metastasis, with metastasis and metastastic site, which was lowest in metastastic site,P=0. 000, P=0. 000. The expression of KiSS-1 in tumor with nerves invasion was significantly lower than that without nerves invasion (P= 0. 000), which kendall t correlated coefficient were 0. 689, P = 0. 004. CONCLUSION: The expression of KiSS-1 has close relation to invasion and metastasis of human pancreatic cancer, and may be involved in regulation of this process.
