摘要
目的研究女贞子多糖(PFLL)是否有抗衰老作用。方法昆明种小鼠60只,随机分为正常对照组,模型组,维生素E(VitE)200mg·kg-1·d-1组,PFLL100,200及400mg·kg-1·d-1组。小鼠颈背部sc5%D-半乳糖1mg·kg-1·d-1,7周,制备衰老模型;正常对照组小鼠sc等量注射用水。同时按分组ig给予相应药物,对照组和模型组ig等量溶剂(只含防腐剂泥泊金),每日1次,给药7周。分别用TBA比色法、亚硝酸盐法、DTNB法及Sohal等法测定心、肝、肾中丙二醛(MDA)含量,超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和脑中脂褐质(LF)含量。结果衰老模型组小鼠胸腺指数和脾脏指数较正常对照组下降,脑组织中LF升高,心、肝、肾组织中MDA含量升高,SOD及GSH-Px活力下降。与模型组比,PFLL和VitE抑制胸腺指数和脾脏指数下降,其量效关系呈正相关;对抗心、肝、肾组织中MDA升高及脑组织中LF升高,其量效关系均呈负相关;抑制心、肝、肾组织中SOD及GSH-Px活力下降,其量效关系均呈正相关。结论PFLL具有抗衰老作用,其机制可能与其增强免疫功能,清除氧自由基和活性氧,提高机体抗氧化酶活力有关。
AIM To study if the polysaccharides from Fructus ligustri lucidi (PFLL)possesses antisenile effect. METHODS Kunming mice were randomly divided into control, model, vitamin E ( Vit E ) 200mg·kg^-1·d^-1, PFLL 100,200 and 400 mg·kg^-1·d^-1groups, 10 mice in each group. Mice were injected sc D-galactose 1 mg·kg^-1·d^-1 for 7 weeks to prepare senile model. At the same time, PFLL 100, 200 and 400 mg·kg^-1·d^-1 or Vit E 200 mg·kg^-1·d^-1 were given ig for 7 weeks. Malondialdehyde ( MDA ) content, superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities in heart, liver and kidney were measured by TBA colorimetric method, nitrite method and DTNB method, respectively, and lipofuscin (LF) content in brain was determined by Sohal method. RESULTS Compared with control, thymus index and spleen index significantly descended; LF content in brain significantly ascended; MDA content significantly rose and SOD and GSH-Px activities significantly dropped in heart, liver and kidney of mice in model group. Compared with model, descending of thymus index, spleen index, SOD activity and GSH-Px activity was dose-dependently inhibited in PFLL groups. Ascending of LF content in brain and MDA content in heart PFLL liver and kidney. CONCLUSION has the notable antisenile effect, its mechanism probably related to improving immunity, eliminate oxygen free radicals and active oxygen and strengthen activity of antioxidase.
