摘要
为探索中药桔梗的主要活性成分桔梗总皂甙及其转化型桔梗皂甙的生物学活性和抗IBDV、A IV(H9)、NDV的作用,采用M TT法和间接免疫荧光检测试验,分别研究桔梗总皂甙与转化型桔梗皂甙的细胞毒性、对CEF生长和活性的影响以及抗病毒活性。结果表明:桔梗总皂甙和转化型桔梗皂甙对细胞毒性作用的IC50分别为128.74和53.48μg.mL-1,对CEF细胞生长和活性具有最大促进作用的浓度分别为80和25μg.mL-1。桔梗总皂甙阻断IBDV、A IV(H9)、NDV感染CEF细胞的作用分别为66.3%、55.4%和36.9%,抑制三者增殖的作用分别为69.4%、60.8%和26.2%;转化型桔梗皂甙阻断三者感染CEF细胞的作用分别为84.7%、63.5%和45.5%,抑制三者增殖的作用分别为77.8%、63.5%和41.9%,表明桔梗总皂甙经化学处理转变成转化型桔梗皂甙,可明显提高其生物学活性和抗病毒用。
To explore the bioactivity of the main active ingredients-platycodon in radix platycodonis and its transformed type, MTT method and indirect IFA were used to study the cell toxicity of platycodon and its effect on the growth of CEF cell line and antivirus activity. The results showed that platycodon and its transformed type had the cell toxicity to CEF cell llne, with the IC50 128- 74μg·mL^-1 and 53. 48μg·mL^-1 respectively, the ideal concentrations for the promotion of growth and cell activities were 80 μg·mL^-1 and 25 μg·mL^-1, respectively. The antivirus activity assay suggested that platycodon could pretect CEF cells infected by IBDV, AIV, NDV, with different inhibition rate 66.3%, 55.4% and 36.9% respectively, platycodon also inhibited IBDV, AIV and NDV to multiplication in CEF cell line by 69.4%, 60. 8% and 26.2% respectively. The transformed platycodon prevented the viruses intruding into CEF ceils by 84.7%, 63.5% and 45.5%, and the multiplication inhibition rates were 77.8%, 63.5%and 41.9% respectively. These results indicated that the chemical modification to platycodon may enhance its bioactivities and the ability of antivirus activity.
