摘要 将芘丁酸(1-pyrenebutyricacid,PBA)通过活性酯法与牛血清白蛋白(BSA)共价结合,以PBA-BSA结合物为免疫原制备抗血清,利用该抗血清建立了测定多环芳烃类物质的间接竞争酶联免疫吸附分析法(enzyme-linkedimmunosorbentassay,ELISA),并研究了10种多环芳烃与该抗血清的交叉反应。结果表明,该方法用于测定芘和芘丁酸的IC50值分别为0.132mg·L-1和0.253mg·L-1,检出限分别为0.1mg·L-1和0.01mg·L-1,该方法不受溶剂甲醇的影响,因此可以用于经过预富集的环境样品中多环芳烃类物质的测定。 1-pyrenebutyric acid (PBA) was conjugated to bovine serum albumin (BSA) by active ester method. Antiserum was raised against the conjugate of PBA-BSA. An indirect competitive enzyme-linked immunosorbent assay (ELISA) for determination of polycyclic aromatic hydrocarbons (PAHs) was proposed by using the obtained antiserum. Cross-reactivities of 10 PAHs with this antiserum were also presented. The IC50 of pyrene and PBA were 0.132 mg·L^-1 and 0.253 mg·L^-1, the detection limits were 0.1 mg·L^-1 and 0.01 mg ·L^-1, respectively. This ELISA was not interfered by methanol and could be used for detection of PAHs in preconcentrated environmental samples.
