摘要
目的:建立高效液相色谱-电化学检测法(HPLC-ECD)测定人尿中8-羟基脱氧鸟嘌呤核苷(8-OHdG)的检测方法。方法:采用Bond Elute C18小柱固相萃取尿液,ODS C18分析柱,8%甲醇醋酸缓冲液为流动相,电化学检测器检测,电极电压750mV。结果:本方法线性范围25~400μg/L,r=0.9892,回收率为91.2%~108.8%,批内和批间精密度分别为3.9%~7.5%和4.3%~8.6%,检测限为2μg/L。结论:本方法操作简便、准确度较好,已应用于正己烷接触者现场尿样的测定。
Objective: To establish a HPLC -ECD method to measure 8- oxo- deoxyguanosine (8 -OHdG) in human urine. Methods:The urinary sample was pretreated with a solid phase extraction (SPE) column. Then the chromatography was performed by an ODS Cls column with ECD detector and mobile phase of acetic acid buffer which contained 8% methanol. Results:The linear range was 25 400 μg/L, r =0. 9892, the recovery rate was 91.2% ~ 108.8%, the intra - and inter - day precisions were 3.9% ~7.5% and 4. 3% ~ 8. 6% respectively, the detection limit was 2 μg/L. Conclusion: It is a better method for the determination of 8 - OHdG in human urine.
