摘要
目的:建立测定全血中秋水仙碱的简便、灵敏、准确的高效液相色谱二极管阵列(DAD)检测方法。方法:全血样品经1%三氟醋酸乙腈溶液提取后,用Waters Oasis MCX小柱进行净化,在ZORBAX Eclipse XDB-C8柱(150mm×2.1mm×5μm)上以乙腈+0.04%三氟醋酸(23+77)为流动相,二极管阵列(DAD)紫外245nm检测,柱温35℃,外标法定量。结果:秋水仙碱在0.50-50.0mg/L范围内均具有良好的线性,血样中定量检出下限为0.10mg/L,方法回收率为88.1%-101.9%,RSD小于10%。结论:该方法的建立具有灵敏度高、操作简便、快速、准确,适用于中毒病人的中毒诊断。
Objective:To establish a simple,sensitive and accurate method for the determination of colchicine in whole blood by HPLC - DAD. Methods: After colchicine in whole blood was extracted by 1% Trifluoroacetic - Acetonitrile, the sample was cleaned with solid phase extraction using Oasis MCX cartridges and the colchicine was separated on ZORBAX Eclipse XDBC8 column( 150 mm × 2. 1 mm ×5 μm) with DAD detection at 245 nm by using the mobile phase consisting of Acetonitrile (23%) and 0. 04% Trifluoroacetic (77%). The column temperature was controlled at 35℃. Results:Calibration curve was linear within the range of 0. 50 - 50. 0 mg/L and the limit of quantification was found to be 0. 10 mg/L The extraction recovery was between 88.1% - 101.9% ,the RSD was less than 10%. Conclusion:This method is sensitive, fast and accurate for determination of trace colchicine in whole blood and can be used in the diagnosis of the clinical poisoned patients.
