一株耐酸硫酸盐还原菌的分离筛选及生理特性研究 被引量:8

摘要 从四川化工集团硫酸厂废水污泥中分离到一株耐酸硫酸盐还原菌.分离菌属于中温菌,最佳生长温度为36℃;轻度耐盐,浓度在1%左右.分离菌具有较宽的pIq生长范围(5.0~8.5),且在pH5.0时的细胞生长量达到最高,与pH7.0时的生长水平相同.相对于已报道的硫酸盐还原菌,具有很强的耐酸性能.分离菌的另一显著特点是碳源利用范围广,能分别利用甲酸钠、乙酸钠、乙醇、丙酸钠、乳酸钠、葡萄糖作为电子供体,进行硫酸盐异化还原.菌体呈杆状或孤状,大小为(0.5~0.6)×(1.7~2.0)肿,革兰氏染色阴性,芽孢染色阴性,单极生鞭毛,做波浪式运动或翻转运动.当培养基中不舍Fe^2+盐时,菌落为白色,直径约1.5mm.当培养基中含有Fe^2+盐时,菌落为黑色,直径约2mm.菌落边缘不整齐,表面较粗糙.根据形态和生理生化特性,该菌株属于脱硫弧菌属(Desulfovibrio). A strain acid-resistant sulate-reducing bacterium was isolated from the sewage of the sulfatefaetory of Siehuan Chemical Industry Group. The strain belongs to mid-the rmophilie strain and its optimum temperature for the growth is 36℃. It is light salt-resistant and the NaC1 coneentration for the growth is about 1% (W/V). The strain can lives in wide pH range (5.0-8.5) and its OD reach the highest when the pH 5.0. This result is same to that when the pH 7.0. It is very aeid-resistant relative to thesulfate-redueing bacterium have been reported. Another obvious charaeteristicof the strain is that it can use many different carbon sources. It can use formate sodium, aeetete sodium, ethanol, pyru rate sodium, laetete soudium and glucose as electrondonor respectively. Its rod or are single cell is about 0. 5-0. 6p.m wide and 1.7-2.0lxra long. The strain is gram negative, doesn't form spore and have a flagellum. It always moves retroflexly or wavily. When the strain grows on the medium without Fe^2+, the colonies are white, 1.5 mm in diameter and on the medium with Fe^2+, the colonies are black and 2 mm in diameter and not smooth. The strain belongs to Desulfovibrio sp based on its modality and physiology.
