摘要
应用随机扩增多态DNA(RAPD)技术分析了湖南、宁夏、黑龙江大体型东方田鼠以及黑龙江小体型东方田鼠4类东方田鼠的基因组DNA。结果发现:4类东方田鼠类群内个体间的相似性都较高,湖南、宁夏、黑龙江大体型东方田鼠以及黑龙江小体型东方田鼠的类群内平均带纹相似系数(ABS)分别为0.9287±0.0436、0.9223±0.0533、0.9515±0.0289、0.8900±0.0328;湖南、宁夏和黑龙江大体型东方田鼠3类鼠之间的遗传相似系数都达0.9以上,遗传距离都小于0.1,黑龙江小体型东方田鼠与其他3类鼠的相似性较低,遗传相似系数在0.6762~0.7203之间,遗传距离在0.3281~0.3913之间;UPMGA聚类分析图显示湖南、宁夏和黑龙江大体型东方田鼠聚为一类,黑龙江小体型东方田鼠单独为一类;引物S2022可以作为区分黑龙江小体型东方田鼠与其他3类东方田鼠的标记引物。
The genome DNA of four groups of Microtus fortis from Hunan, Ningxia and Heilongjiang were analyzed by the random amplified polymorphic DNA (RAPD) technique. The RAPD results revealed that: The average band similarity index (ABS)within each group of Microtus fortis is high: for voles from Hunan, 0.9287±0.0436; for voles from Ningxia, 0.9223±0.0533; for big voles from Heilongjiang, 0.9515±0.0289; for small voles from Heilongjiang, 0.8900±0.0328, The genetic similarity coefficients (I) among the voles from Hunan, Ningxia and the big voles from Heilongjiang were all up to 0.9 and the corresponding genetic distances (D) of them all under 0.1. But the genetic similarity coefficients between the small voles from Heilongjiang and the other groups were ranged from 0.6762 to 0.7203 and the genetie distances between them ranged from 0.3281 to 0.3913. According to the clustering analysis of the genetic distance using UPGMA, the voles from Hunan, Ningxia and the big voles from Heilongjiang could be gathered in one group, but the small voles from Heilongjiang were belonged in the other group alone. The primer S2022 could be used as the characteristic primer to distinguish the small voles from Heilongjiang from the other three groups of Microtus fortis for it could produce two specific DNA markers.
