结缔组织生长因子体外促进人增生性瘢痕成纤维细胞转分化的研究 被引量:17

摘要 目的研究结缔组织生长因子(connectivetissuegrowthfactor,CTGF)在人增生性瘢痕成纤维细胞转分化中的作用。方法培养人增生性瘢痕成纤维细胞,分别给予不同剂量CTGF刺激(10ng/ml),以及CTGFASODN转染,48h后用Westernblot方法比较各组间α平滑肌肌动蛋白(αsmoothmuscleactin,αSMA)的表达变化。结果刺激48h后,对照组表达少量αSMA蛋白;小剂量CTGF刺激组和大剂量CTGF刺激组作用48h后,αSMA表达量均显著增多,且成浓度依赖性变化(P<0.01);而CTGFASODN转染组αSMA蛋白表达与对照组比较明显减少(P<0.05)。结论CTGF在体外能促进人增生性瘢痕成纤维细胞向肌成纤维细胞(myofibroblast,MyoF)的转分化,阻断或者抑制其表达可能将更有效的治疗瘢痕挛缩。 Objective To investigate the role of connective tissue growth factor (CTGF) in transdifferentiation of fibroblasts from human hypertrophic scar (HSF). Methods The fibroblasts from hypertrophic scar were passaged and then divided into 4 groups according to stimulation factors: stimulated with 10% FBS-DMEM ( negative control) , with 2.5 ng/ml rhCTGF, with 10 ng/ml rhCTGF, transfected with CTGF ASODN. The expression of α-smooth muscle actin(α-SMA) was measured by Western blotting. Results The expression of α- SMA in fibroblasts increased markedly after 2.5 ng/ml or 10 ng,/ml rhCTGF treatment (P 〈0.01 ), while decreased markedly after CTGF ASODN transfection (P 〈 0.05 ). Conclusion CTGF can promote the transdifferentiation of fibroblasts of human hypertrophic scar towards myofibroblasts in vitro. Inhibition of CTGF expression may be a more effective therapy of scar contracture.
