摘要
采用50对SSR特异引物对甜瓜栽培品种(系)进行分析,其中48对扩增出谱带,46对引物具有多样性。扩增带分子量在250 ̄1750bp之间,187条谱带中有130条谱带具有多态性(占69.52%),平均每个引物可扩增出3.729条带。46份材料经过NTSYS软件分析后分为9组,从9组中再选取21个具有代表性材料重新进行聚类,应用NTSYS软件的立体旋转模拟分析和聚类分析两种方法。结果表明,21份材料分为8组,15份材料具有相同的分析结果,6份材料具有不同的分析结果。聚类结果显示21份材料可分为绿麻瓜类、黄白皮瓜类、白皮瓜类、面瓜类、菜瓜类等几个类型,基本与形态学分类相似,但也有特殊性,如花皮面瓜聚到绿麻瓜一类中。分析结果说明这两种方法具有各自的特点,要根据研究目的来选择和确定分析方法。此外,运用分子标记的方法可以提高甜瓜栽培品种多样性分析的准确性。
The genetic diversity among 46 Chinese melon cultivars (Cucumis melo L.) from different area in China was assessed by variation of 50 simple sequence repeat primers. It resulted that there were total 46 polymorphic bands on 48 amplified bands using fifty primers of simple sequence repeat. Total 187 alleles, which 130 of them had polymorphism, revealed the average of 3.729 alleles per marker, and the molecular size was between 250-1 750 bp. 46 Chinese melon cultivars were divided into nine groups using NTSYS program and methods of multidimensional scaling and cluster tree analysis. 21 characterization melon accessions from 9 different groups were selected to analysis again, they were divided into 8 groups. There were the same results on 15 melon accessions, 6 melon accessions had different conclusion after compared two methods of MDS and cluster tree analysis by NTSYS program. It showed difference between two analyses, and two methods had advantage respectively. Moreover, these date indicated that genetic diversity of Chinese melon cultivars could be enhanced by the introduction of genetic variation from different origins of Chinese accessions, at the same time it would be advantageous to aequire more accessions from this geographically varied region to ensure the retention of existing genetic diversity. SSR markers play an important role in identification and study of genetic diversity.
