泌乳素腺瘤中PTTG的表达与肿瘤侵袭性关系 被引量:2

摘要 目的探讨泌乳素腺瘤中垂体瘤转化基因(p itu itary tumor transform ing gene,PTTG)与肿瘤侵袭性生长的关系。方法手术切除的泌乳素腺瘤标本46例(侵袭性24例和非侵袭性22例),正常垂体组织6例(对照),采用免疫组化方法测定PTTG蛋白表达;从组织中提取RNA,经RT-PCR方法扩增PTTG mRNA;从细胞中提取蛋白,行W estern b lot检测。结果PTTG蛋白和PTTG mRNA表达在侵袭组和非侵袭组差异显著,并且通过免疫组化和W estern b lot两种方法检测PTTG蛋白表达水平相一致。结论PTTG与肿瘤的侵袭性密切相关,对肿瘤的发生、发展有重要作用,可以作为临床判定肿瘤侵袭生长的分子生物学标志。 Purpose To explore the relationship between pituitary tumor transforming gene(PTTG) expression in human prolactinoma and invasive growth of the tumor.Methods Forty-six cases of prolactinoma were divided into the invasive group and the non-invasive group.Expression of PTTG protein was examined by immunocytochemical method.Total RNA was extracted from tissue specimens and PTTG mRNA was detected by RT-PCR;expression of PTTG protein was detected by Western blot.Results Expression of PTTG protein and PTTG mRNA in human prolactinoma was significantly different between the invasive group and the non-invasive group.Expression level of PTTG protein by immunocytochemical method was consistent with that of PTTG protein by Western blot.Conclusions The expression of PTTG is correlated with invasiveness of the tumor,which may play an important role in pathogenesis and development of prolactinoma.It can serve as a molecular marker for invasive growth in human prolactinoma.
