用两种不对称催化反应合成紫杉醇C_(13)侧链

摘要 目的合成紫杉醇C13侧链(2R,3S)-3-苯甲酰胺基-2-羟基-3-苯基丙酸乙酯;方法合成一种新的可回收的小分子手性配体,分别催化不对称氨羟化和二羟化反应合成紫杉醇C13侧链;结果均获得高光学纯度的目标产物,化学产率39%和49%,光学纯度97%和99%;结论两种方法中各步反应产物纯化方法简单,配体均可回收和重复使用。 OBJECTIVE To develop a simple and economical method to achieve taxol side chain, (2R, 3S)-isoproplyl-3- benzamido-2-hydroxy-3-phenylpropionate. METHODS The taxol C13 side chain with high ees could be prepared from asymmetric aminohydroxylation (AA) and dihydroxylafion (AD). RESULTS Total yield of C13 side chain was 39% and 52% , the enantiomertie excess was 97% and 99%. CONCLUSION The two synthetic manipulations are simple and practical, the ligand can be easily recovered and reused.
