摘要
以小麦条锈菌夏孢子为实验材料,分析比较了CTAB法、SDS法、尿素法和CTAB/SDS法4种方法对小麦条锈菌基因组DNA的提取效果。结果表明,利用4种方法提取的小麦条锈菌夏孢子DNA的纯度和得率存在一定差异,单位重量夏孢子分离的DNA量依次为:CTAB法>尿素法>CTAB/SDS法>SDS法,而所获得的DNA纯度依次为:CTAB/SDS法>CTAB法>SDS法>尿素法。以不同分离方法获得的小麦条锈菌DNA为模版,利用特异性微卫星引物RJ17进行PCR扩增和电泳分析,4种方法均能满足SSR反应,但CTAB法更适于SSR反应。
Uredospores of wheat yellow rust was used as test material, isolation methods of wheat yellow rust genomic DNA by CTAB, SDS, Urea and CTAB/SDS were compared.The results showed that the purity and yield of wheat yellow rust DNA obtained by above methods were different.These four methods were ranked as: CTAB〉Urea〉CTAB/SDS〉SDS according to the average DNA microgram of uredospores, and ranked as: CTAB/SDS〉CTAB〉SDS〉Urea according to the genomic DNA purity.The DNA obtained by these four methods was subjected to SSR analyses with R J17 primer ,and the DNA obtained with CTAB method used as template was better than others in SSR analysis.
