Determination of Excitatory Amino Acid in the Cerebral Tissue of Neonatal Rat with Hypoxia-ischemia and the Effect of Dexamethasone by Capillary Electrophoresis

摘要 建立了新生儿缺氧缺血性脑病(H IE)发生时脑组织中兴奋性氨基酸(EAA)含量的高效毛细管电泳-激光诱导荧光检测(CE-LIF)的新方法。方法的最佳条件为:40 mmol/L硼砂溶液,分离电压20 kV,压力进样5 s,λem=488 nm、λex=560 nm。随机分为假手术组、新生儿缺氧缺血性脑病(H IE)组,小剂量地塞米松(DXM)组和大剂量DXM组的新生大鼠脑组织中EAA的含量测定,结果表明:假手术组EAA含量较低,H IE组EAA含量显著升高,大、小剂量DXM预处理后,EAA含量较H IE组有不同程度的降低。 A method of (CE-LIF) has been dev high performance capillary electrophoresis with doped for the determination of the concentration laser-induced fluorescence detection of the excitatory amino acid (EAA) in the cerebral tissue of neonatal rat with hypoxia-ischemic encephalopathy (HIE). The effects of several factors such as the acidity and concentration of running buffer, the separation voltage, the applied potential and the injection time on CE-LIF were investigated. Under the optimum condition, the three amino acids L- glutamic acid, L-asparic acid and glycine can be separated in 40 mmol/L borax running buffer, 20 kV separation voltage,5 s injection time, λem = 488 nm, λex = 560 nm. The results from the concentration of shameful operation group, HIE group treated with hypoxia-ischemia without pretherapy, small dose dexamethasone (DXM) group and large dose DXM group showed hypoxia-ischemia could increase the releases of EAA, the pretreatment of large dose DXM prior to hypoxia-ischemia is effective in protecting brain ceils from further hypoxia-ischemia damage by reducing the EAA's releases. The method is simple, accurate, easy-to-operate, and the assay results are satisfactory.
