摘要
目的:探讨外源性VEGF诱导的大鼠视网膜血管病变的特点。方法:大鼠120只随机分为正常对照组、rrVEGF164低、高浓度组。不同时间行荧光素眼底血管造影、视网膜电图振荡电位、组织学检查。结果:高浓度组注射rrVEGF164眼从注射后10d表现为眼底后极部视网膜动脉局限性高荧光,2wk时表现为单个视网膜内核层毛细血管管腔窄小,内皮细胞肥大,管腔面积和细胞面积分别与自身对照眼、4,6wk比较,差异有显著性(P<0.05)。视网膜电图OPs总波幅2,6wk降低,分别与自身对照眼、4wk比较,差异有显著性(P<0.05)。结论:高浓度高频率注射rrVEGF164诱导的大鼠视网膜血管病变主要表现有视网膜水肿,大血管扩张,内核层毛细血管管腔变窄、接近闭塞,内皮细胞肥大、增生,周边部玻璃体视网膜纤维血管样膜。
AIM: To investigate characteristics of retinal vasculopathy induced by exogenous VEGF in rats.
METHODS: A total of 120 rats were randomly divided into three groups: normal control group, low concentration rrVEGF164 group and high concentration rrvEGF164 group. Fundus fluorescein angiography, ERG- OPS and histological observation were performed in every group on schedule.
RESULTS: Limited hyperfluorescence appeared at the central of retinal dilated vessels in posterior pole 10d after the first intravitreal injection. The individual retinal capillary lumen of the inner nuclear layer constricted, retinal capillary endothelial cells hypertrophy at 2wk in the high concentration rrVEGF164 group. Compared with self-controlled eyes and the experimental eyes 4wk and 6wk after the first intravitreal injection respectively, Significant differences were found (P〈0.05). Total amplitudes of retinal OPs in high concentration rrVEGF164 eyes decreased at 2wk and 6wk. Significant differences were found, compared with self-controlled eyes and the experimental eyes at 4wk (P〈0.05).
CONCLUSION: Retinal vasculopathy induced by high concentration rrVEGF164 with high frequency in rats is characterized by retinal edema, vessels dilation, constricted and nearly closed capillary lumen of the inner nuclear layer, retinal capillary endothelial cells hypertrophy and proliferation in inner nuclear layer, firbrovascular-like membrane in the peripheral retina.
