摘要
目的:检测海马CA1,CA3区域的钙离子荧光强度的变化,观察钙超载的情况,并观察亚低温对癫痫发作的影响,方法:实验于2003-03/08在华中科技大学同济医学院药理学实验室进行。①实验分为两部分:一部分14只大鼠用于钙超载的检测,其中7只用做致痫模型(癫痫组),7只用作对照组,另取5只用作尼莫地平的阻断试验(药物组)。另一部分28只用于观察不同温度致痫大鼠行为学和组织病理学改变。②用荧光倒置显微镜来测定癫痫状态下海马CA1,CA3区钙超载的情况,并用钙离子拮抗剂尼莫地平作用于海马脑片观察钙超载的情况。③28只大鼠分为头皮温度为41℃,37℃,32℃,26℃4组,通过对4组点燃大鼠行为学和海马组织病理学的变化观察亚低温对癫痫的脑保护作用。结果:47只大鼠均进入结果分析。①癫痫大鼠海马CA1,CA3区钙超载高于对照组,CA3区钙超载高于CA1区,在尼莫地平的作用下CA3区钙超载明显降低。②亚低温下癫痫的发作程度最轻,高温下癫痫的发作程度最重,随温度降低发作程度也递减。③不同温度海马组织学改变从重至轻依次为41℃组>37℃组>26℃组>32℃组。亚低温下CA3区神经元的丢失最少,温度越高,CA3区神经元的丢失越严重,温度越低,CA3区神经元的丢失也越轻。结论:钙超载参与了癫痫的发病,其中海马CA3区的钙超载在癫痫发病中起主要作用,而亚低温对脑有保护作用。
AIM: To detect the change of fluorescence intensity of calcium ion in hippocampal CAI and CA3 region and observe the condition of calcium overload and the effect of mild hypothermy on epileptic attack.
METHODS: The experiment was carried out at the laboratory of pharmacology, Tongji Medical College, Huazhong University of Science and Technology between March and August 2003. ①The experiment was assigned into two parts: the 14 rats were used in the detection of calcium overload, in which 7 rats were used for inducing epilepsy model (epilepsy group), and 7 rats were used as control group, and other 5 rats were used as the nimodipine blockage trial (drug group). The 28 rats of the second part were used to observe the change of behavior and histopathology of rats in different temperature. ②The status of calcium overload at hippocampus CA1 and CA3 regions under epilepsia status was detected with fluorescence inverted microscope. After the calcium ion antagon nimedipine affected on hippocampal brain slice, the status of calcium overload was observed. ③Totally 28 rats were assigned into 41 ℃, 37 ℃, 32 ℃, 26 ℃ scalp temperature groups. The cerebra] protective effect of mild hypothermy on epileptic was observed by the changes of behavior of kindling rats and histopathology of hippocampal tissues of the 4 groups.
RESULTS: Totally 47 rats were involved in the result analysis. ①The calcium overload at hippocampi CA1 and CA3 regions of epilepsia rats was higher than that of the control group, and that in CA3 region was higher than that of CAI region. Under the influence of nimedipine, the calcium overload at CA3 region decreased significantly. ②Epileptic episode degree was light in mild hypothermy, while in high temperature it was the severest, and decreased with the decrease of temperature. ③The change of hippocampal histology was 41 ℃ group 〉 37 ℃ group 〉 26 ℃ group 〉 32 ℃ group from severe to mild in order. The lose of neurons at CA3 region in mild hypothermy was the minimal. The higher the temperature, the severer the lose of neurons at CA3 region was, while the lower the temperature, the milder the lose of neurons at CA3 region was.
CONCLUSION: Calcium overloading is involved in the onset of epilepsies, while calcium overloading in hippocarnpi CA3 region plays the main role in the onset of epilepsies. While mild hypothermla has protective effect on brain.
