三品系果蝇的低氧半数致死时间和低氧预适应的保护作用 被引量:1

Fifty Percent Lethal Time of Anoxia and Protective Effect of Anoxia Preconditioning in Three Strains of Drosophila melanogaster

摘要 目的观察对七氟醚敏感S、耐药R,野生H三品系果蝇的低氧半数致死时间(LT50)和低氧预适应后存活率的改变。方法240只成年S或R品系果蝇随机分入低氧(氧浓度<0.03%)3.0、3.54、.0、4.5、5.0或5.5h;H品系则随机分入低氧2.0、2.5、3.03、.5、4.0或4.5h,6组,每组40只。Bliss法计算LT50。另各品系80只成年果蝇随机分入低氧预适应组和对照组,每组40只。72h后观察两组果蝇的存活率。结果S、R品系LT50比H品系明显延长(P<0.01,n=8),S、R品系间则无差异(P>0.05,n=8)。R品系果蝇低氧预适应组存活率比对照组增加14.01%(P<0.05,n=12);H、S品系则无明显变化。结论S、R品系果蝇比H品系更耐低氧,S、R品系相似。三品系中,仅R品系存在低氧预适应保护作用。 Objective S, H and R, three strains of fruit flies were used as an experimental animal to investigate their 50% lethal time ( LT50 ) of anoxia and their changes of survival probability after anoxia preconditioning. Methods 240 fruit flies from S or R strain were randomly exposed to anoxia ( below 0.03 % oxygen ) for periods of 3.0,3.5,4.0,4.5,5.0 or 5.5 h, and H strain for periods of 2.0,2.5,3.0,3.5,4.0 or 4.5 h. LT50 of anoxia in the three strains was calculated with the method of Bliss. Another 80 fruit flies from each strain were randomly exposed to anoxia preconditioning or air (the control group) treatment. The survival probability was observed in three strains between the two groups. Results LT50 of S strain or R strain was significantly longer than that of H strain ( P 〈 0.01, n = 8 ), and there was no significant difference between S strain and R strain ( P 〉 0.05, n = 8). The survival probability of R strain receiving anoxia preconditioning was increased by 14.01 ( P 〈 0.05, n = 12) ,but not in H or S strain. Conclusion S and R strains of fruit flies are similar tolerance to anoxia but mere tolerant to anxia than that of H strain. Through anoxia preconditioning treatment, only R strain shows a protective effect.
