高氧对早产新生大鼠肺组织细胞色素b、三磷酸腺苷合成酶6表达的影响 被引量:8

摘要 目的探讨高浓度氧暴露对早产新生大鼠肺组织细胞色素b(Cyt b)、ATP合成酶6(ATPase 6)动态表达的影响。方法早产新生SD大鼠生后1 d随机分为空气组、高氧组。高氧组持续暴露于常压氧舱中,氧质量浓度>85%;空气组置于同一室常压空气中。两组分别于高氧或空气暴露后1、4、7、101、4 d提取肺组织RNA,采取半定量逆转录聚合酶链反应(RT-PCR)测定Cyt b、ATPase 6 mRNA表达;应用免疫组化方法检测肺组织切片Cyt b。结果1.与空气组比较,高氧暴露14、d Cyt b mRNA含量及其表达显著增强(P<0.05);7 d后Cyt b呈下降趋势,其表达较空气组减弱,但两者比较无显著性差异(P>0.05)。2.高氧组与空气组比较,ATPase 6 mRNA表达于1 d时显著增强(P<0.05),4 d虽无显著改变(P>0.05)但仍增强;7、10 d时减弱;14 d又极显著增强(P<0.01)。3.随日龄变化高氧暴露后,肺组织Cyt b免疫组化结果与Cyt b mRNA表达相似。与空气组比较,高氧暴露14、d肺组织Cyt b表达显著增强(P<0.05);7 d后Cyt b呈渐下降趋势,其表达较空气组减弱,但7、10 d两者比较无显著性差异,14 d极显著减弱。结论线粒体呼吸链中Cyt b、ATPase 6在肺发育中起重要作用。高氧诱导线粒体呼吸链中Cyt b、AT-Pase 6结构和功能改变,导致氧化磷酸化解耦联,参与高氧肺损伤的发病机制。 Objective To explore the effect of hype.roxia on the expression of cytochrome b(Cyt b) and adenosine triphc)sphate synthase 6(ATPase 6) in premature newborn rats' Lung. Methods One - day old preterm SD rats were randomly into air group and hyperoxia group, Hyperoxia group were continuously exposed to oxygen (oxygen 2〉 0.85 )and air group in room air. After 1,4,7,10,14 days of exposure, the rats were killed and these lung RNA was isolated and Cyt b and ATPase 6 mRNA were detected by reverse transcription polymerase chain reaction (RT PCR). Immnohistochemistry was used to detect Cyt b in lung scctions. Results 1. Compared with air group, Cyt b mRNA expression significantly increased (P 〈 0.05) after 1,4 days of exposure; the general tendency decreased after 7 days, and its expression became weak compared with air group, but both were not much different ( P 〉 0, 05 ). 2, Compared with air group, AT- Pase 6 mRNA expression were significantly increased (P〈0.05)after one clay exposure;it did not change significantly by the 4^th day (P〉0.05) ,but increased, And it decreased by 7 and 10 days; by the 14^th day ATPase 6 mRNA expression increased significantly( P〈 0.01 ), 3, With the variation of time, lung sections Cyt b immunohistochemistry results were in accord with Cyt b mRNA expression, Lung sections Cyt b significantly increased ( P〈0.05 )after 1,4 days of exposure; the general tendency decreased after 7 days; its expression became weak compared with air group, but both were not significantly different ( P 〉0.05 ). By 14 days Cyt b expression significantly increased ( P 〈 0.01 ). Conclusions Cyt b and ATPase 6 of mitochondrial respiration chain have vital effects on lung development. Hyperoxia might induce the changes of structure and function of Cyt h and ATPase 6, which could reauh in uncoupling of oxidative phosphorylation (OXPHOS),and play a part role in the mechanism of hyperoxia- induced lung injury.
