摘要
目的:探讨乳腺浸润性导管癌中表皮钙粘蛋白E-cadherin(E-cad)、细胞周期素D1(cyclinD1)、Ki-67蛋白的表达及其临床病理意义。方法:用免疫组织化学EnVision二步法分别检测63例乳腺浸润性导管癌(IDC)组织(IDC组)及15例乳腺纤维腺瘤及15例乳腺小叶增生症乳腺组织(对照组)中E-cad、cyclinD1、Ki-67蛋白的表达情况,结果:E-cad在IDC组及对照组中表达阳性率分别为58.3%、80%;cyclinD1在IDC组及对照组中表达阳性率分别为68.25%、0;Ki-67在IDC组及对照组中表达阳性率分别为65.08%、0。2组间差异均有统计学意义(P <0.05)。E-cadherin、cyclinD1、Ki-67的表达与患者年龄及肿块大小无关,E-cadherin、cyclinD1的表达与组织学分级、淋巴结转移相关。结论:E-cad、cyclinD1、Ki-67表达在乳腺癌发生、发展演进过程中是一个重要指标。联合检测E-cadherin、cyclinD1、Ki-67蛋白对预测乳腺癌发生、发展及淋巴结转移有重要意义。
Objective: To explore the clinicopathologic significance of the protein expression of E-cadherin (E-cad), cell cycle regulator cyclin D1, as well as proliferative cell nuclear antigen ki-67 in invasive ductual carcinoma of the breast. Methods: Immunohistochemical EnVision method was used to detect the expres- sion of E-cad, cyclin D1 and Ki-67protein in 63 cases with IDC, 15 cases with adenoma and 15 cases with adenosis. Results: The positive rates of E-cadherin protein expression were 58.73% in IDC, 80%in adenoma and adenosis, respectively. The positve rates of cyclin D1 protein expression were 68.25% in IDC, 0 in adenoma and adenosis, respectively. The positive rates of Ki-67 was 65.08% and 0 between the two group, respectively. There were significant difference between the two groups in the expression of E-cad, cyclin D1, Ki-67 (P 〈0.05). At the same time,the expression of E-cad and cyclin D1 protein was correlated to lymph node metastasis in IDC (P 〈0. 05). The expression of E-cad and cyclin D1 was correlated to the grade of tissue science. But there was no correlation between expression of E-cad, cyclin D1 and Ki-67with tumor size and patient's age. Conclusions: The experssion of E-cad,cyclin D1, Ki-67 is an important marker during the process of origination and development in breast carcinomas. The combined detection of E-cad, cyclin D1 ,Ki-67 has positive effects on predicting the orignation and development and the lymph node metastasis in breast carcinomas.
