摘要
目的:观察克汀病模型大鼠下丘脑神经递质去甲肾上腺素,5-羟色胺,5-羟吲哚乙酸对促甲状腺激素代谢的影响。方法:实验于2004-08/12在锦州医学院地甲病研究所完成。取SD雄性大鼠20只,单纯随机分为模型组和对照组2组(n=10)。模型组用8g/LNaClO4饮水复制克汀病大鼠,对照组饮用自来水,各组每周称重,观察大鼠行为状态。3个月后测体质量后处死,迅速断头取血,制备血清,测定三碘甲状腺原氨酸、甲状腺素和促甲状腺激素水平;低温下取脑及甲状腺,用荧光法测定下丘脑5-羟色胺、5-羟吲哚乙酸和去甲肾上腺素的含量,同时测甲状腺相对质量及含碘量,然后进行促甲状腺激素与上述下丘脑神经递质间的相关性分析。结果:20只大鼠全部进入结果分析。①模型组大鼠甲状腺肿大,生长发育、运动明显迟缓。②模型组大鼠体质量和甲状腺含碘量明显低于对照组(P<0.005),而甲状腺相对质量明显高于对照组(P<0.005)。③模型组大鼠下丘脑5-羟色胺、5-羟吲哚乙酸、去甲肾上腺素和5-羟吲哚乙酸/5-羟色胺比值均高于对照组(P<0.05,0.01)。④模型组大鼠血清三碘甲状腺原氨酸、甲状腺素浓度低于对照组,促甲状腺激素水平高于对照组(P<0.01)。⑤去甲肾上腺素、5-羟吲哚乙酸、5-羟吲哚乙酸/5-羟色胺与血清促甲状腺激素均呈正相关(r=0.68,0.81,0.76,P<0.01)。结论:大鼠下丘脑去甲肾上腺素及5-羟色胺系统能促进促甲状腺激素的分泌和释放,下丘脑组织中5-羟色胺系统在甲状腺功能低下条件下代谢比较活跃,这种神经递质代谢的变化可能是克汀病某些异常行为的生化基础。
AIM: To observe the influences of hypothalamus norepinephrine (NE), 5- hydroxytryptamine (5-HT) and 5-hyaroxyindoeacetic acid (5-HIAA) on the metabolism of thyroid stimulating hormone (TSH) in rats with cretinism.
METHODS: The experiment was completed in the Institute of Endemic Goiter, Jinzhou Medical University from August to December 2004. Twenty male SD rats were divided into two groups at random: model group (n=10) and control group (n=10), Rat models of cretinism were duplicated by drinking 8 g/L NaC104 in the model group, and the rats in the control group drank tap-water, all the rats were weighed every week, and the behavioral status was observed, The body mass was measured and then the rats were killed at 3 months, head was quickly cut to prepare serum, and then the levels of triiodothyronine, tetraiodothyronine and TSH. The brain and thyroid gland were removed under hypothermia, and the contents of 5-HT, 5-HIAA and NE were determined by means of fluorescence, at the same time, the relative mass of thyroid gland and content of iodine were detected, and then the correlation between TSH and the above-mentioned hypothalamus neurotransmitters was analyzed.
RESULTS: All the 20 rats were involved in the analysis of results. ①Thyromegaly, and delay of growth and development and motion were observed in the rats of the model group. ② The body mass and content of iodine in thyroid gland of rats in the model group were obviously lower than those in the control group (P 〈 0.005), but the relative mass of thyroid gland was markedly higher than that in the control group (P 〈 0.005).③The contents of hypothalamus 5-HT, 5-HIAA and NE and the ratio of 5-HIAA to 5-HT in the model group were all higher than those in the control group (P 〈 0.05, 0.01).④The serum levels of triiodothyronine and tetraiodoth- yronine in the model group were lower than those in the control group, but the level of TSH was higher than that in the control group (P 〈 0.01). ⑤ The levels of NE, 5-HIAA and ratio of 5-HIAA to 5-HT were positively correlated with serum level of TSH (r=0.68, 0.81, 0.76, P 〈 0.01).
CONCLUSION: NE and 5-HT system of hypothalamus can boost secretion and release of TSH. 5-HT metabolism of hypothalamus on hypothyroidism was active and changes of the neurotransmitter may be the biochemical basis of some abnormal behavior in patients with cretinism.
