目的观察脂多糖(LPS)对大鼠垂体前叶滤泡星形细胞分泌S-100b蛋白的影响。方法将原代培养大鼠垂体前叶细胞分为对照组和LPS处理组,经LPS不同剂量处理后以作者科室建立的敏感和特异的S-100蛋白ELISA检测方法测定培养上清中和细胞裂解液中S-100b蛋白水平。结果大鼠垂体前叶细胞培养上清中S-100b蛋白水平随细胞生长过程逐渐增高,尤其在培养前12 h增高明显。培养上清中乳酸脱氢酶水平无明显变化。LPS可剂量依赖性地抑制培养上清中S-100b蛋白水平增高。在培养12 h时,对照组细胞的胞浆中S-100b蛋白水平显著增加,LPS可完全抑制胞浆中S-100b蛋白水平上升。结论LPS抑制大鼠垂体前叶滤泡星形细胞分泌S-100b蛋白,提示感染时LPS可能通过改变S-100b蛋白分泌而影响垂体前叶细胞的功能。
Objective To observe the effects of lipopolysaccharide (LPS) on the release of S-100b protein from cultured rat folliculo-stellate cells (FSC) in adenohypophysis. Methods Primary cultures of rat adenohypophysis cells were treated with either LPS or vehicle control, followed by measurements of S-100b protein contents in cell lysate and in extracellular media using a highly specific and sensitive a ELlSA kit developed in our laboratory. Results S-100b protein concentrations in culture medium gradually increased with the duration of culture, which became most pronounced during the initial 12 hours of culture. In contrast, no change was seen in LDH activity in culture supernatant. LPS dose-dependently prevented the gradual increase in S-100b protein contents in medium. In addition, elevations in intracellular levels of S-100b protein were also abolished by incubation with LPS. Conclusions LPS inhibits the release of S-100b protein from rat FSCs. The results suggest that, during infection, LPS in blood could alter anterior hormone release by affecting the release of S-100b protein, a highly possible mediator of plciotropic actions of FSC.
