摘要
目的从樟树花粉中克隆泛变应原基因。方法通过对多个植物泛变应原基因进行序列同源性比对,根据保守区域序列设计简并性引物,利用RT-PCR结合RACE技术对樟树花粉中的泛变应原基因进行克隆。结果从樟树花粉中克隆到一个新的基因。序列分析显示:所克隆到的基因与其它植物的泛变应原肌动蛋白结合蛋白基因有很高的同源性,因此认为该基因为泛致敏原基因,命名为Cinc1。其在GenBank数据库中的登录号为DQ335252。结论采用简并引物从樟树花粉中克隆到一泛变应原基因,为进一步研究该变应原奠定了理论基础。
Objective To clone new panallergen genes from pollen of C Sequences of panallergen gene from various species were compared and the conservative domains among the sequences were used for degenerate primer designing. RT-PCR and RACE methods were applied to clone the fur length panallergen gene from pollen of Cinamomum camphora. Results A new cDNA clone was obtained. Sequence analysis showed that this clone shared a high sequence homology with panallergen profilin from other species. The gene, designated as Cin c 1, was regarded as a new panallergen profilin gene. The C, enBank accession number for this clone was DQ335252. Conclusion A new panallergen gene Cin c 1 was cloned from pollen of Cinnamonmm camphora using degenerate primer. Further study of Cin c 1 may enhance our understanding of this panallergen.
