摘要 目的探讨X射线照射后鼻咽癌CNE-1细胞产生多药耐药的机制.方法采用MTr法检测X射线照射后CNE-1细胞对顺铂(cD-DP)的耐药性;采用逆转录-PCR(RT-PCR)和蛋白免疫印迹(Western blot)方法,检测照射后CNE-1细胞mdrl基因的表达.结果随照射后时间的延长,cDDP对细胞的半数抑制浓度(IC50)值逐渐增大,第2~4周IC50值显著高于照射前(P<0.01).照射后细胞mdrl mRNA和P-gp的表达较照射前均显著增强(P<0.01).结论X射线照射可诱导CNE-1细胞mdd基因的表达,这可能是照射后肿瘤细胞产生多药耐药的重要机制之一. Objective To explore mechanism of multi-drug resistance in nasopharyngeal carcinoma (NPC) CNE-1 cells after X-ray irradiation. Methods The drug resistance of CNE-1 cells after X-ray irradiation to cisplatin (cDDP) was determined by MTY. The expression of multidrug resistance ( mdr 1 ) gene was tested by RT-PCR and Western blot. Results ICS0 values of cDDP in cells 2 ~ 4 weeks after irradiation were signiticantly higher than those before irradiation (P 〈0.01 ). The expression of mdrl mRNA and P-gp in cells after irradiation increased than those before irradiation (P 〈 0.01 ). Conclusions X-ray irradiation may induce the expression of mdrl gene in CNE-1 cells, which maybe play an important role in muhidrug resistance after NPC irradiation.
