摘要
用快速酶联免疫吸附试验双抗体夹心法(RDS-ELISA),测定了6株抗日本乙型脑炎病毒(JEV)单克隆抗体(nG2、2H4、2D2、mG9、2F2、mC3)的抗原结合位点及相对亲和力。通过ELISA相加试验证实,6株单克隆抗体识别5个不同的抗原位点.2H4和2F2识别的抗原位点相同或接近,而其余单克隆抗体则识别不同的或相距较远的抗原位点。从50%最大结合的单克隆抗体浓度分析可知,6株单克隆抗体的相对亲和力为nG2>2H4>2D2>mG9>2F2>mC3,浓度范围约为0.01~1.50ng。本研究为JEV单克隆抗体在诊断学及其疫苗研究中的应用提供了必要的资料。
On the basis of rapid double sandwish enzyme-linked immunoassay (RDS-ELISA), we determined the antigenic sites and relative affinity of the six anti-Japanese encephalitis virus ( JEV ) monoclonal antibodies (McAb). It was found by ELISA additivity test that, the six McAb recognized five different antigenic sites.2H4 and 2F2 recognized the same or very similar antigenic site, but the antigenic sites recognized by the other McAb were different. By analysing the 1/2 maximum binding of the McAb, the relative affinity of the McAb could be ranked as following. nG2>2H4 >2D2>mG9>2F2>mC3, at the protein concentration of 0.01 to 1.50 ng. This study is useful for the applications of the JEV-McAb in immunologi-cal diagnosis and vaccine preparation.
