蝎毒多肽Ⅲ和蛇毒多肽Ⅷ对小鼠Lewis肺癌生长和肺部转移的抑制作用

摘要 目的:探讨蝎毒多肽Ⅲ(PSV)和蛇毒多肽Ⅷ(KD)对Lewis肺癌(LLC)荷瘤小鼠肿瘤生长、肺部转移和一般状态的影响。方法:采用近交系C57BL/6小鼠,左腋下接种LLC细胞悬液建立肺转移模型,随机分为 5组,每天分别腹腔注射生理盐水(NS)、环磷酰胺(CTX)、高剂量PSV(PSV-H)、低剂量PSV(PSV-L)和KD,共 18 d。以原位瘤重、肺转移率和转移数为指标,检测不同浓度的PSV和KD对LLC生长和转移的抑制作用;以体重和脾系数为指标观察PSV和KD对带瘤小鼠一般状态的影响。结果:结果显示,PSV和KD对LLC原位肿瘤的抑制作用不明显(P>0.05),但它们对肿瘤肺转移却有非常明显的抑制作用PSV-H 0.33±0.2l,(P< 0.01);其免疫保护方面的作用也明显优于CTX。结论:蝎毒多肽Ⅲ和蛇毒多肽Ⅷ抑制原位瘤的作用缓慢,但是抑制肿瘤转移方面强于化疗药物CTX,并能够提高小鼠的生存质量。 Objective:To observe the effects of scorpion-derived peptide PSV and snake venom peptide KD on proliferation and metastasis of carcinoma cells and general conditions in Lewis Lung Carcinoma (EEC)-bearing mice. Method: Thirty-eight inbred C57BL/6 female mice were inoculated with LLC cells (5.0 × 106) in left armpit subcutaneously and then divided into 5 groups randomly to receive one of the following: normal saline (NS), cyclophosphamidum (CTX), high or low-dose PSV or KD intraperitoneally once a day for 18 days. Weight of tumor, numbers and rates of tumor metastasis to the lung were used to evaluate the inhibitory effects of PSV and KD on the proliferation and metastasis of LLC cells. The body-weight and spleen weight index were observed to study the effects of PSV and KD on general conditions of the LLC-bearing mice. Results : Inhibitory effect of PSV and KD on tumor weight was not significant compared with positive controls (CTX) ( P 〉 0.05 ). Numbers and rates of tumor metastasis to the lung were decreased obviously in both PSV and KD treated groups. Compared witll CTX treated group, the difference was significant ( P 〈 0. 05 ). And PSV and KD can protect the immunity system of the tumor-bearing mice. Conclusion: PSV and KD showed weak effect on tumor proliferation, but provided better protection from metastasis than conventional chemotherapy and improved the life quality of tumor-bearing mice.
