摘要
目的观察p53在db/db糖尿病小鼠颌下腺中的表达及细胞凋亡。方法选取3、4、6、8、10月龄db/db糖尿病小鼠及相应月龄的db/+m正常小鼠颌下腺,应用SP免疫组织化学方法及TUNEL标记方法染色后进行图象分析,统计p53及凋亡细胞在颌下腺组织中表达及分布的细胞阳性率。结果①p53及凋亡细胞在对照组及糖尿病组颌下腺中均有表达及分布,糖尿病组细胞阳性率均高于对照组。②糖尿病组p53及凋亡细胞阳性率随月龄增大均呈增加趋势。结论①p53的表达随月龄增大上升,提示随着糖尿病发展,颌下腺实质细胞有凋亡趋势。②凋亡细胞阳性率随糖尿病发展而增加显著,这与糖尿病时腺体的逐渐萎缩和功能受损相一致。
Objective To observe the expression of p53 and disitribution of apoptotic cells in submandibular gland of db/db mutant diabetic mice. Methods Fifty mice were randomly divided into 2 groups, db/db diabetic group and control group. The expression of p53 and disitribution of apoptotic cells were observed by using and TUNEL. Results results were as foUow:① p53 was expressed in submandibular gland from both diabetic mice and control mice.②The percentage of immunopositive cells of p53 was significandy more in diabetic mice than those of control mice, and there was continuous enhance of p53 as disease progressed in diabetic groups. TUNEL results were as follow: ①Apoptotic cells were founded in submandibular gland in both diabetic mice and control mice. ②The percentage of apoptotic cells was significant increased in diabetic mice than those of control mice, and showed enhance in both groups. Conclesion ① The percentage of immunopositive cells of p53 were increased with development of the disease, which could associate with the dysfunction of epithelial cells. ②The enhance of apoptotic cells demonstrates that diabetes mellitus can induce apoptcsis, which may be the reason of the submandibular gland atrophy.
