益活清胰Ⅰ号对大鼠重症急性胰腺炎心肌损伤的保护作用 被引量:5

摘要 目的研究益活清胰Ⅰ号对重症急性胰腺炎(thesevereacutepancreatitis,SAP)大鼠心肌损伤的保护作用,探讨其作用的机制。方法63只SD大鼠随机分为治疗组(n=27)、SAP模型对照组(n=27)和假手术组(n=9),SAP模型采用5%牛磺胆酸钠胰胆管逆行注射方法建立。ELISA法检测血清CTn1、CK-MB水平,免疫组化法检测心肌组织NF-κB的表达,光镜及电镜下观察胰腺及心肌组织的病理变化。结果大鼠SAP造模术后6h开始出现CTn1、CK-MB水平升高,治疗组各时点CTn1、CK-MB水平较对照组明显下降(P<0.01)。SAP大鼠各时间点均可见心肌组织NF-κB明显活化,治疗组各时间点NF-κB活化与对照组相比明显减弱(P<0.01)。光镜及电镜下可见SAP大鼠的心肌组织存在明显的变性、坏死等病理损伤,治疗组胰腺及心肌组织病理损伤明显减轻。结论益活清胰Ⅰ号能抑制NF-κB在心肌组织中的活化,减轻SAP大鼠心肌的损伤,对SAP合并心脏损害的具有保护作用。 [Objective] To investigate the effect of the Chinese herbs, No.1 Yihuo Qingyi on severe acute pancreatitis (SAP) and SAP-associated myocardial damage in rats. [Method] Sixty-three male SD rats were divided randomly into the sham-operated group (A group, n=9), the control group (B group, n=27) and the Chinese herbs treatment group (C group, n=27). SAP was induced by intraductal administration 5% sodium taurocholate. Artery blood of the A, B, C group was obtained at 6 h, 12 h and 18 h after induction of the model respectively. Serum concentrations of CK-MB and CTnl were detected by ELISA. The expression of NF-κB in myocardial tissue was determined by SP immunohistochemical technique. Pancreatic and myocardial tissues were examined under light and electron microscopy. [Result] The contents of CTnl, CK-MB in serum was significantly increased in 6 h after induction of SAP, the contents of CTnl and CK-MB in serum were significantly decreased, compared with those of the control group (P 〈0.01). NF-κB activity in myocardial tissue was significantly increased in 6 h after induction of SAP, and myocardial NF-κB activity of the treatment group was significantly decreased compared with those of the control group (P 〈0.01). Pancreatic and myocardial tissues of the treatment group of the pathological changes was alleviated in contrast to the control group. [Conclusion] Promote Blood Circulation Qing Yi Decoction can inhibit the activity of NF-κB in the myocardial tissue, alleviate the pathological damage of SAP rats and myocardial damage following SAP .Promote Blood Circulation Qing Yi Decoction is a potential effective medicine for SAP therapy.
