摘要
为研究细胞在不同生理状态下某些必需微量元素组成及含量的变化,应用同步辐射 X 荧光测定细胞在衰老、缺氧和外源性自由基损伤时 Fe、Zn 含量的变化。并探讨其相互关系。3组实验结果显示相同规律,即衰老、缺氧和外源性自由基作用都使细胞过氧化程度增高,表现为脂质过氧化的分解产物丙二醛(MDA)量增加,细胞内 Zn 含量显著减少,Fe/Zn比值同步增大。在补充 Zn 或金属硫蛋白后,MDA 量下降,Zn 含量增加,Fe/Zn 比值趋于正常。实验结果表明,细胞内微量元素 Fe、Zn 含量和比值随细胞不同生理状态而变化。同步辐射 X 荧光作生物样品微量元素定量分析比带电粒子荧光分析更为优越。
PURPOSE To investigate the changes on composition、concentration of some essential trace elements in cells un- der various physiological and pathological status.METHODS Synchrotron radiation X-ray fluorescences (SR-XRF) were applied to analyse the change of cellular Fe、Zn contents and their interrelation in aged and anoxic cells,as well as injured cells by free rad- icals.RESULTS In all three groups malondialdehyde (MDA) content was raised together with the Fe/Zn ratio.Zinc or metal- lothionein (MT),could inhibit lipoperoxidation by decreasing MDA and Fe/Zn ratio.CONCLUSIONS Iron,zinc and their ratio vary in accordance with the physiological status of the cell.SR-XRF is a powerful tool for the quantitative analysis of trace ele- ments in samples of cells.
