摘要
目的观察锌缺乏与锌过量对小鼠胚胎肢芽细胞分化的影响及其毒性作用。方法取12天龄昆明种小鼠胚胎的肢芽,应用微团培养技术,观察不同剂量的锌对肢芽细胞分化和增殖的影响。结果培养基中锌缺乏时或者培养基锌浓度达42μmol/L时,均可明显抑制细胞分化和增殖(P<0.01)。当培养基锌浓度为21μmol/L或28μmol/L时,对肢芽细胞分化和增殖均具有明显的促进作用,而锌浓度达35μmol/L时,对细胞分化的促进作用转化为抑制作用但不影响细胞的存活。结论体外实验条件下,锌对小鼠胚胎肢芽细胞的分化和增殖表现为促进和抑制双重作用;缺锌及过量锌均可抑制细胞的分化并产生毒性作用。
Objective To investigate the potential influence of zinc deficiency and excessiveness on the differentiation and cytotoxicity to mice embryo limb bud cells. Method The micromass culture system of limb bud cells from Kunming mice embryos at 12 - day stage was used to evaluate the cytotoxicity of zinc and its effect on embryonic chondrogensis in vitro. Results Zinc deficiency appeared to be cytotoxic to mouse embryo limb bud cells in culture and inhibited the clone formation. As amount of ZnSO4 ·7H20 added to 21 μmol/L and 28 μmol/L, the cell differentiation and proliferation could be promoted, but when the amount of zinc added excessively above 35 μmol/L, both of the proliferation and the proliferation were affected. Conclusions The data suggested that different amounts of zinc might affect not only the proliferation but also the proliferation on mouse limb bud cells in vitro. Excessive zinc could significantly induce cytotoxicity and inhibit cell differentiation.
