摘要
目的探索应用组织工程技术建立新型稳定的瘢痕成纤维细胞三维培养模型的方法。方法在瘢痕成纤维细胞单层培养的基础上,应用新型高孔隙率海绵状聚乳酸-乙醇酸(PLGA)支架为成纤维细胞培养载体,建立成纤维细胞三维培养体系。应用冰冻切片及扫描电镜观察瘢痕成纤维细胞三维培养的生长特性。结果在此三维培养模型中,细胞、细胞外基质以及各种调控因素共同构成一个有机整体,其中成纤维细胞所表达的生物学特性更接近于它们在体内瘢痕组织中的特点。结论本实验应用组织工程技术成功地建立了新型瘢痕成纤维细胞三维培养体系,研究成纤维细胞的各种生物学特性,是研究创伤愈合和瘢痕形成领域里一个新的和更为科学的方法。
Objective To study the methods of establishing a new stable tissu engineering scar fibroblast three-dimensional culture system. Methods Base on the monolayer cell culture system, using PLGA as a substrate, a new three-dimensional cell culture system was built. The biologic characteristics of the fibroblasts were observed by means of freezing microtomy and scanning electron microscopy. Results In this culture model, cell, extracellular matrices and various modulating factors were associated to form an organic entity,in which the fibroblasts expressed their biologic characteristics similar to that in living body. Conclnsion A new tissue engineering scar fibroblasts three-dimensional cell culture system was developed, the system ought to recommend as a better model in the research of wound healing and scar formation.
