摘要
吸水链霉菌NND-52发酵液的上清液经乙酸乙酯萃取,硅胶柱粗分离,经制备型高效液相色谱仪分离纯化,得除莠霉素A(herbimycinA)、黄色M3纯品和浅黄色M4粗品,粗品结晶后获得M4的白色针状结晶;通过基本理化性质和光谱分析,确定M3、M4分别为除莠霉素C(herbimycinC)、双氢除莠霉素A;噻唑蓝(MTT)法检测除莠霉素A、除莠霉素C及双氢除莠霉素A对Vero、B16-F10细胞增殖的影响,对肿瘤细胞活性强度顺序为:除莠霉素C>除莠霉素A>双氢除莠霉素A。除莠霉素C可能是一种更具潜力的抗肿瘤抗生素。
Antibiotic M3 and M4 were isolated from the cultured broth of Streptomyces hygroscopicus NND-52 by ethyl acetate extraction, silica gel column chromatography and preparative high performance liquid chromatography. Basic physical and chemical characteristics and spectra data showed antibiotic M3 and M4 are herbimycin C and dihydroherbimycin A, respectively. The effects of herbimycin A, herbimycin C and dihydroherbimycin A on proliferation of Vero and B16-F10 cells were investigated by MTT assay. Results showed that their antitumor activities were as follows: herbimycin C〉herbimycin A〉dihydroherbimycin A. Compared with herbimycin A, herbimycin C is a more potentially applicable antitumor antibiotic.
