摘要
目的通过与 UW 液进行比较.探讨 SWH 液对 SD 大鼠肝脏保存的有效时间。方法应用大鼠原位肝移植模型,通过高效液相色谱法(HPLC),检测肝组织 ATP、ADP、AMP,计算总腺苷酸含量(TAN)及 Atkinson's 能量转换(EC),判断肝脏能量代谢状态;通过检测谷丙转氨酶(ALT)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)含量,判定肝脏功能;观察移植术后7d 动物存活率,判定 SWH 液对大鼠肝脏有效保存的时间。结果保存18 h 组的 ATP、TAN、EC 恢复水平明显低于保存8 h 组,相差显著。保存12 h 组与保存8 h 组比较相差不显著;保存18 h 组的 ALT、AST、LDH明显高于保存8h 组.差异显著,保存12 h 组与保存8 h 组比较相差不显著;SWH 液组移植术后7 d动物存活率有高于 UW 液组的趋势。结论无论使用 SWH 液还是 UW 液,SD 大鼠肝脏有效保存的时间都不超过12h。
Objective To determine the effective time of storage of rat liver in SWH solution and compare the solution with UW solution. Methods After the model of orthotopic liver transplantation was established in rats, the levels of ATP, ADP and AMP of hepatic tissue were determined by HPLC. Meanwhile, the levels of ALT, AST and LDH were measured and the animal 7-d survival rate determined. Results The levels of ATP, TAN and EC were significantly lower in the 18h group than in 8h group. No significant differences were found between the 12h group and 8h group. The levels of AST, ALT and LDH were markedly increased after 18h of preservation than 8h of preservation. There were no remarkable differences between the 12h group and 8h group. The animal 7-d survival rate was markedly higher in SWH group than in UW group. Conclusions The liver of SD rats should not be stored over 12 h with SWH solution or UW solution.
