摘要
目的观察在高浓度葡萄糖条件下,体外培养兔视网膜Müller细胞碱性成纤维生长因子(basic fibroblastgrowth factor;bFGF)的表达及其钠(Na+)通道的变化。方法采用免疫细胞化学,透射电镜方法鉴定体外培养的兔视网膜Müller细胞;应用免疫细胞化学方法定性观察高糖条件下视网膜Müller细胞bFGF表达的变化;利用膜片钳技术观察高糖条件下Müller细胞Na+通道的变化。结果高糖可以刺激视网膜Müller细胞表达bFGF,使Na+通道的开放概率降低。结论高糖刺激Müller细胞bFGF的表达,而发挥促血管生成的作用,在此过程中Na+通道的开放概率降低。
Objective To observe the effects of high concentration glucose on bFGF expression and sodium channels of rabbit retinal Müller ceils cultured in vitro.Methods Transmission electron microscopy and immunocytochemical staining method were used to identify Müller cells. The changes of bFGF expressed by Müller cells were studied with immunocytochemical method in a qualitative way. Patch-clamp technique was used to observe the changes of sodium channels caused by high concentration glucose. Results bFGF expression was enhanced obviously by high concentration glucose, which can inhibit sodium channels of rabbits' Müller cells cultured in vitro. Conclusion bFGF expression secreted by cultured Müller cells in vitro can be enhanced at high concentration glucose. Müller cells play a potential role in neovascularization of diabetic retinopathy. High concentration glucose can inhibit sodium channels during the process.
