多孔淀粉接枝聚合物载体固定化纤维素酶的研究 被引量:11

摘要 合成了淀粉接枝丙烯腈、丙烯酰胺两亲性高分子化合物,并以此为载体,共价偶联固定纤维素酶。探讨了加酶量、反应体系pH值、偶联时间等因素对固定化酶的影响,并对固定化酶的性质进行了研究,结果表明:最适温度为50℃,最适pH值为6.6,热稳定性、重复使用稳定性和储存稳定性均有明显提高。 Cellulase was immobilized onto starch-g-poly(acrylonitrile/acrylamide) copolymers carrier by means of diazotization. The effects of different coupling time, buffer pH and the amount of cellulase on activity and specific activity of immobilized enzyme were studied. Some characteristics of immobilized and free cellulases, such as optimum temperature, optimum pH, stabilities to heat, the operational stabilities and the stabilities for reservation were studied and compared. The studies on property of immobilized cellulase showed that the optimal pH is 6.6 and the optimal temperature is 50℃. The thermal stability and the stability for reservation of immobilized cellulase are improved considerably.
