摘要
目的探讨去整合素 Kistrin(disintegrin kistrin)对体外培养的人眼晶状体上皮细胞(human lensepithelial cells,HLECs)增殖的抑制作用。方法对先天性白内障患者术中环行撕前囊膜,进行原代培养并传代,取第二代细胞用于实验。增殖抑制实验:将悬浮的活细胞接种于涂有鼠尾胶原的96孔培养板,16 h 后,去掉旧培养液,换用不同浓度的去整合素(5ng/ml、10ng/ml、20ng/ml、40ng/ml、80ng/ml)37℃、5%CO_2条件下共同孵育,7d后,MTT 法测定晶状体上皮细胞的数目。结果 20ng/ml、40ng/ml、80ng/ml 的去整合素对体外培养的晶状体上皮细胞的增殖进行干预后,OD 值分别为0.254±0.008、0.224±0.009,0.202±0.010,与空白对照组相比,差异有统计学意义(P<0.05)。结论去整合素可抑制体外培养的人眼晶状体上皮细胞的增殖。(中国眼耳鼻喉科杂志,2006,6:146~148)
Purpose To investigate the inhibitory effect of disintegrin Kistrin on the proliferation of human lens epithelial cells(HLECs) in vitro. Methods In the congenital cataract surgery, the anterior capsule membrane sampled by CCC technique was cultured and subcultured and the second generations of HLECs were used in this experiment. Antiproliferative experiment: 16 hr after the second passage of HLECs was incubated in the 96-well plate coated with rat-tail collagen at 37℃ ,5% CO2, they were exposed to disintegrin Kistrin with different concentrations (5ng/ml,10 ng/ml,20 ng/ ml,40ng/ml,80 ng/ml). Seven days later, the number of cells was determined by MTT method. Results Compared with the control group, the OD values of HLECs incubated in disintegrin Kistrin, at the concentrations of 20 ng/ml,40 ng/ml and 80 ng/ml for 7 days(0.254 ± 0.008,0.224 ± 0.009 and 0.202 ± 0.010 respectively) were decreased significantly (0.266 ± 0.08.5 in control group, P 〈 0.05). Conclusion In vitro, disintegrin Kistrin plays a significant role in inhibiting the proliferation of HLECs.
