摘要
以酰氯A对消旋体α-苯乙胺进行柱前衍生化处理,以正己烷和异丙醇混合体系为流动相,在自制的手性柱(DNB-PG CSP)上对其进行了对映体分离研究,考察了流动相配比对分离因子的影响,研究发现正己烷与异丙醇的体积比为98∶2时,取得了理想的分离效果,分离因子α值达1.26.
In order to research the possibility of the enantioseparation of - α - methylbenzylamine ( α - PEA) by HPLC. The compound of one sort of acyl chloride A was used as the derivating reagent. And then the acematic α- PEA derivative was resolved on DNB - PG chiral stationary phase with the mobile phase of Hexane - isopropanol system. The effect of mobile phase compositions on the resolution selectivity was investigated. The satisfactory separation effect with α value up to 1.26 has been found when the 98:2 (volume ratio) Hexane - isopropanol being taken as the mobile phase. And the possible chiral recognition mechanism in the progress of enantiomer separation was also discussed in this article. The result indicated that the derivative of α - PEA could be resolved very well under the given chromatography condition.
