益肾降浊汤对大鼠脑缺血再灌注损伤脑组织抗氧化酶活力的干预效应 被引量:2

Interventional effect of Yishen Jiangzhuo decoction on activity of antioxidase in brain tissues of rats after cerebral ischemical reperfusion injury

摘要 目的:观察脑缺血再灌注后大鼠脑组织丙二醛、钙离子含量和超氧化物歧化酶活性的变化,探讨益肾降浊汤对其损伤脑组织抗氧化酶活力的影响。方法:实验于2003-04/2004-09在承德医学院基础医学研究所进行。采用Wister大鼠120只,雌雄各半,随机分为5组,每组24只,即正常对照,假手术组,模型组,益肾降浊汤组,都可喜组。益肾降浊汤由泽泻、何首乌、黄连、肉苁蓉、石菖蒲、三七组成,由承德市药材公司提供,水煎、过滤、浓缩至每毫升药液含生药2.53g;都可喜(阿米三嗪与萝巴新按一定比例的混合制剂),法国施维雅药厂生产,上海福达制药有限公司分装(批号:90841)。对模型组,益肾降浊汤组,都可喜组在造成大鼠高脂血症的基础上,进行脑缺血再灌注。都可喜组按20mg/kg都可喜,益肾降浊汤组按2.53g/kg生药,灌胃给药,均为1次/d,从术前3d开始至杀检为止。假手术组,高脂血症大鼠仅做双侧颈总动脉分离术,然后缝合,不进行缺血再灌注,正常对照组:不加任何处理。各组分别于造模后第1,7,15天断头取脑,每时间点8只。用羟胺法测超氧化物歧化酶活性,TBA法测丙二醛含量,用原子分光光度法测钙离子含量。结果:实验大鼠120只全部进入结果分析。①第1,7,15天模型大鼠超氧化物歧化酶活性与正常大鼠相比明显降低眼穴16.05±3.17雪,(16.33±1.97),(17.01±4.03),(22.11±2.29)NU/mg;P<0.05~0.01演;而益肾降浊汤组酶活性与同期模型组相比,显著升高眼(18.89±2.36),(20.36±2.46),(22.98±2.14)NU/mg;P<0.05~0.01演。②模型组大鼠3个时间点脑组织内丙二醛含量显著高于正常对照组眼(11.78±1.32),(10.15±1.44),(8.15±2.16),(8.56±1.46)μmol/g,P<0.05~0.01演,而益肾降浊汤组3个时间点的丙二醛含量较同日期模型组明显降低眼(11.78±1.32),(10.15±1.44),(8.15±2.16)μmol/g,P<0.05~0.01演。③各组钙离子含量与丙二醛含量呈现相同的变化趋势。结论:益肾降浊汤可提高脑组织内超氧化物歧化酶活性,降低缺血再灌后脑组织丙二醛含量及钙离子含量,能增加脑缺血再灌注模型大鼠脑组织抗氧化酶的活性,保护脑神经元。 AIM: To observe the changes of contents of malondialdehyde (MDA) and calcium ion as well as activity of superoxide dismutase (SOD) in brain tissue of rats after cerebral ischemical repeffusion injury, and explore the effect of Yishen Jiangzhuo decoction on activity of antioxidase of injured brain tissue. METHODS: The experiment was performed at the Institute of Basic Medicine, Chengde Medical College from April 2003 to September 2004. Totally 120 Wister rats, male and female (half and half), were selected and randomly assigned into 5 groups with 24 rats in each group: normal control group, sham operation group, model group, Yishen Jiangzhuo decoction group and duxil group. The Yishen Jiangzhuo decoction was composed of alisma rhizoma, fleeceflower root, coptidis rhizoma, caulis cistanchis, acori graminei and panax ,pseudoginseng, provided by Chengde Municipal Medical Material Company. This decoction was decocted in water, filtered, and concentrated till 2.53 g crude drug in per milliliter physic liquor, Duxil (mixture preparation of almitrine and raubasine according to a certain proportion) was produced by France Les Laboratoires Servier, packed fractionally by Shanghai Forward Pharmaceutical Co,. Ltd. (batch number: 90841), The rats in the model group, Yishen Jiangzhuo decoction group and the duxil group received cerebral ischemical reperfusion injury based on the onset of hyperlipemia, The rats in the duxil group received 20 mg/kg duxil and those in the Yishen Jiangzhuo decoction group received 2.53 g/kg crude drug by gastric perfusion, once a day from 3 days before operation to kill. The rats with hyperlipemia in the sham operation group only underwent commissurotomy at bilateral common carotid artery, and then sutured, but not underwent ischemical reperfusion injury, The rats in the normal control group did not undergo any treatment, The rats in each group were killed to gain brains at the 1^st, 7^th and 15^th days, respectively, 8 rats in each time point, Activity of SOD was determined with hydroxylamine method, the content of MDA was detected with TBA method, and the content of calcium ion was measured with atomic spectrophotometric method. RESULTS: A total of 120 experimental rats were involved in the result analysis. ①The activity of SOD of model rats significantly decreased at the 1^st, 7^th and 15^th days as compared with the normal rats [(16.05±3.17), ( 16.33 ± 1.97 ), ( 17.01 ±4.03 ), (22.11 ±2.29 ) NU/mg ;P 〈 0.05-0.01], The activity of SOD of the rats in the Yishen Jiangzhuo decoction group obviously increased as compare with the model group in the same period [(18.89±2.36), (20.36±2.46), (22.98±2.14) NU/mg;P 〈 0.05-0.01]. ② The content of MDA of rats in the model group was markedly higher than that in the normal control group at the 3 time points [(11.78±1.32), (10.15±1.44), (8.15±2.16), (8.56±1.46) μmol/g,P 〈 0.05-0.01], while the content of MDA in the Yishen Jiangzhuo decoction group remarkably decreased as compared with the model group in the same period [(11.78±1.32), ( 10.15±1.44), (8.15±2.16) μmol/g,P 〈 0.05-0.01]. ③The changes of calcium ion content was the same to that of the MDA content in each group. CONCLUSION: The Yishen Jiangzhuo decoction Can elevate the activity of SOD of brain tissue, decline tbe contents of MDA and calcium ion of brain tissue after ischemical reperfusion injury, accelerate the activity of antioxidase of brain tissue of rats after cerebral ischemical reperfusion injury and protect cerebral neurons.
