番荔枝总提取物对二乙基亚硝胺诱发大鼠肝脏癌前病变的影响 被引量:3

摘要 目的探讨番荔枝总提取物对二乙基亚硝胺(DEN)诱发大鼠肝癌的防治作用。方法按改良的SoltFarber氏方法制作大鼠肝脏癌前病变模型,利用免疫组化、血清和组织生化等检测方法,观察番荔枝总提取物对DEN诱发的肝癌形成的影响。结果番荔枝总提取物与提取物的二氯甲烷部位各组大鼠肝表面癌结节数、肝/体重比、肝组织匀浆和血清中谷胱甘肽-S-转移酶含量,以及血清γ-谷氨酰转移酶、碱性磷酸酶的含量明显低于模型组。免疫组化显示,模型组大鼠的肝脏组织中bax免疫组化表达很少,bcl-2免疫组化表达较多;而番荔枝总提取物组bax免疫组化表达较多,bcl-2免疫组化表达较少。结论番荔枝总提取物能抑制癌前病变,延缓DEN诱发大鼠肝癌的发展,但不能阻断DEN诱发大鼠肝癌的发生。 Objective To investigate the effect of Annona glabra extract (AGE) in preventing DEN-induced Hepatocarcinogenesis in Rats. Methods The modified Slot-Farber precancerous liver rat model was used.Immunohistochemistry,serum and tissue biochemistry methods were used to study the effects of AGE on the development of DEN-induced hepatocarcinoma of rats.Results After 18 weeks,the number of cancerous node of the AGE groups including the Extract from the leaves and the part of dichlormethane from the seeds was significantly lower than that in the DEN group,the ratios of liver/body weight were lower than those in the model group,the content of GST in liver homogenate supernatant and serum γ-GT,AKP and GST levels in the AGE groups were significantly lower than those in the DEN group respectively.The bcl-2 gene expression in the liver tissue of the AGE groups was lower than those in the model group, contrarily;the bax gene expression of the AGE groups was higher than those in the model group. Conclusion AGE can protect liver cells form damage induced by DEN and the devel opment of hepatoma may be delayed, but it cannot inhibit production of hepatoma completely.
